چکیده:
The aim of this study by performing thedevelopment of gadolinium(III) oxide(Gd (2) O( 3) ) nanoparticleswith diethylene glycol polymer (DEG) and also by achievementof the contrast enhancement evaluation of Gd loadednanoparticles in comparison with Magnevist(Gd-DTPA), wasto indicate that Gd (2)O(3)nanoparticles with diethylene glycolpolymer could produce a good MR signal and therefore couldbe a useful potential contrast medium for cell tracking inmagnetic resonance molecular imaging(MRMI).This study would be complicated with nanoparticles composedgadolinium(III) oxide (Gd2O3) with diethylene glycol polymer.The size and morphological structure of this Nano particledetermined by particle size analysis device(zeta sizer) andTransmission Electronic Microscope . Proton relaxation timeswere measured with a 1.5-T MRI scanner. The measurementswere performed in aqueous solution.Other purpose of this study was to assess cytotoxicity ofgadolinium oxide nanoparticles. The effects of nanoparticleson U-87 MG, THP_1 and SK_MEL 3 cell lines were evaluatedby light microscopy, and by standard cytotoxicity assays.The results showed a significantly higher incrementalrelaxivity for Gd(2)O(3) nanoparticles compared to Gd-DTPA.The slope of R1 relaxivity(1/T1) vs. concentration curve of Gd-DTPA and Gd (2)O(3)were 4.33, 13.37s(-1)mM(-1). The slope of R2relaxivity(1/T2) vs. concentration curve of Gd-DTPA andGd(2)O(3)were 5.06, 9.05s (-1)mM (-1) . Viability results indicate thatU-87 MG, THP_1 and SK_MEL 3 cells endure treatment withGd2O3nanoparticles for an wide-ranging stage of time and it istherefore concluded that results in this study are founded onviable cells. The study indicates the possibility of obtaininghigh relaxivity compared to Gd–DTPA using Gd2O3 ascontrast agent.
